    Autophagic flux chloroquine

Three forms of autophagy are commonly described: macroautophagy, microautophagy, and chaperone-mediated autophagy (CMA). In macroautophagy, expendable cytoplasmic constituents are targeted and isolated from the rest of the cell within a double-membraned vesicle known as an autophagosome, which, in time, fuses with an available lysosome, bringing its specialty process of waste management and disposal; and eventually the contents of the vesicle (now called an autolysosome) are degraded and recycled.

Inhibition of autophagic flux using chloroquine prevented clearance of p62 aggregates, leading to caspase-8 activation and cell death in C4-2 cells. In PC3 cells, inhibition of autophagy induction prevented p62 accumulation and hence caspase-8 activation. A HOS-EGFP-LC3 cells were starved of amino acids or chloroquine CQ treated for 4 hours or left untreated. EGFP fluorescence was measured by FACS either without top or with bottom a wash with PBS containing 0.05% saponin. In the extreme case of starvation, the breakdown of cellular components promotes cellular survival by maintaining cellular energy levels. In disease, autophagy has been seen as an adaptive response to stress, promoting survival of the cell; but in other cases it appears to promote cell death and morbidity.

  6. Chloroquine inhibits the fusion of the autophagosome to the lysosome, thereby permitting the quantitation of the autophagosome formation as a measure of the degree of autophagy by arresting autophagic flux before the lysosomal degradation can occur 18.

Chloroquine is commonly used to study the role of endosomal acidification in cellular processes 2, 3, such as the signaling of intracellular TLRs. Moreover, Chloroquine inhibits autophagy as it raises the lysosomal pH, which leads to inhibition of both fusion of autophagosome with lysosome and lysosomal protein degradation 4. The increased resistance of metastatic cells could be explained by the contribution of the autophagic process as a survival mechanism. Inhibition of autophagy using chloroquine CQ together with LUT treatment in MET4 cells decreased viability significantly more than addition of LUT alone. Ing the basal autophagic flux by decreasing autophagosome-lysosome fusion, and not by inhibiting lysosomal degradation capacity as BafA 1 does. Although treatments with both CQ and BafA 1 ultimately cause a block of the autophagic flux, we show that the intracellular changes that are caused by these 2 compounds are profoundly different.

It has been reported as an side effect of Plaquenil - hair loss or bleaching of the hair, if you think its too bothersome, please ask your doc/pharmacist to prescribe some med which is more comfortable for your system. The Palquenil can't cure the Sjgrens, and the Lyrica takes care of most of my pain. No one seems to want to prescribe certain pain meds for fear of addiction. If I take after my mom, I'll have bald spots by the time I'm in my 70's... Just found out it could be from the hydroxychloroquine. I was told Accutane doesn’t cause hair loss 10 years ago, and now it’s listed as a side effect. Inhibition of Endosomal/Lysosomal Degradation Increases. Inhibitors of endosomal acidification fall into three groups based on their modes of action. The first class of agents is made up of the lysosomotropic weak bases, such as chloroquine, amantadine, and ammonium chloride, which diffuse across membranes in a concentration-dependent manner.
