    Pharmacokinetics of chloroquine in rats

Chloroquine has been extensively used in mass drug administrations, which may have contributed to the emergence and spread of resistance. It is recommended to check if chloroquine is still effective in the region prior to using it.

Abstract. Effect of Gnetum africana on the pharmacokinetic parameters of chloroquine was investigated. Chloroquine phosphate 200 mg/Kg was concurrently administered to overnight fasted albino rats. Blood samples were collected 15, 30, 60, 120, 240 and 480 minutes after administration through cardiac puncture. Chloroquine is a medication used to prevent and to treat malaria in areas where malaria is known to be sensitive to its effects. Certain types of malaria, resistant strains, and complicated cases typically require different or additional medication. Data for allometric scaling were obtained from 6 animal studies mice, rats, rabbits, monkeys. Pharmacokinetics of chloroquine in Thais plasma and red-cell concentrations following an intravenous infusion to healthy subjects and patients with Plasmodium vivax malaria.

The Centers for Disease Control and Prevention recommend against treatment of malaria with chloroquine alone due to more effective combinations. In areas where resistance is present, other antimalarials, such as mefloquine or atovaquone, may be used instead.

  7. Jul 31, 2012 Although chloroquine is a widely used antimalarial and no toxicity was expected at the prophylactic doses used in this study, it was necessary to evaluate the toxicity of the combined treatment. coDbait was administered three times per week during 4 weeks at dose levels of 8, 16, or 32 mg per animal per day in chloroquine-treated rats and monkeys.

Sixty albino Wistar rats were used in the in vivo study of the effects of ethanolic leaf extract of Lasianthera africana on the pharmacokinetic parameters of chloroquine. After an overnight fast, the rats were divided into two groups designated A and B. Group A was given chloroquine only 15 mg/kg and group B received chloroquine 15 mg/ml and L. africanaextract 200 mg/ml concurrently. Reported that V. amygdalina affected the pharmacokinetics of Chloroquine in rat. Hence, there is need to investigate the effect of Vernonia amygdalina on the pharmacokinetics of Dihydroartemisinin.

Hydroxychloroquine plaquenil withdrawl? - LUPUS UK I have read about people stopping P & flaring up. Those who stopped it abruptly had real problems similar to what you are experiencing. If you really need to get off it, I would taper down slowly, very slowly. There is a similar posting on Healthunlocked, "Why oh why did u stop taking plaquenil." You may want to check it out.
